Saw this on YouTube and grabbed a screen shot. Was curious as to what would it would take to build something like this? What body is available in SA? What rolling chassis would one use? V8 petrol or Diesel? Would make an awesome project car?

Thats pretty much a modern jeep chassis with a classic body on.
We do a lot like that - you buy anything built as a body on chassis - Defender, Cruiser, Wrangler - for as much as you want, take the body off, cut the floor out of the classic, make a new floor and trim up. It's the quick and easy way (as opposed to custom building a chassis and suspension.
Discoveries are a good option - D2 or D1 as they are inexpensive and easily modded. The drive train will also accept a modern V8 with a bit more go if you want, or a bigger diesel engine

Lots of bodies in Cape Town too I see. Could put a Morris Minor on a Wrangler. that would be interesting.

it's all possible. We just did one with monster truck tyres and a boat body. Can't post pics of that yet as it's not released, but here is another one on a disco 2 chassis, with a custom body

What do you guys do with them when done? What would a car like that cost to build and register on the road? Thinking of a practical marketing vehicle like the Red Bull ones.

Depends.
Some get kept by the production company.
Some we are made to destroy (this is a legal thing - if they sell to to someone and they later harm themselves, the company can be sued in the USA)
Some get recycled
Some get sold
The ones in the post above were sold into a props house for a very low price, which then went bust and they were auctioned off at even lower prices.
- This one was sold to an Afrikaburn patron
this one is up for sale - hopefully some burners will club together to buy it, or it's a cheap marketing tool for someone.
If you want a custom built vehicle , price very much depends on spec, but it's not a problem to be road legal with the correct bits.
